How to Use TM Media’s MS Medium

Using Murashige & Skoog Medium is as straightforward as it is effective. To prepare the medium, follow these simple steps:

  1. Begin by dissolving 4.10 grams of dehydrated medium in 600 ml of distilled or deionized water at room temperature (15-30°C).
  2. Rinse the media vial with a small quantity of distilled water to ensure no traces of powder remain.
  3. Add any desired heat-stable supplements before autoclaving and stir until the powder has fully dissolved. Sometimes, the medium may not dissolve completely unless the pH is reduced. Lower the pH to about 3.0 to facilitate dissolution. Adjust the medium's pH using buffers.
  4. Make up the final volume to 1000 ml with distilled water, gently mix, and heat. Rotate between intervals until the solution becomes clear. Avoid boiling, reheat, and allow it to cool below 50°C during dispensing.
  5. Dispense the medium into suitable containers, seal or cap them, and autoclave at 15 psi (121°C) for 15 minutes, using a slow exhaust cycle. Do not recommend higher temperatures or longer times.
  6. Cool the autoclaved culture vessels containing medium to 45-50°C and aseptically add any desired sterile heat-labile substrate.
